Occasionally when I'm working in an assembly the mate "paperclip" button gets disabled (greyed). I don't know what I did to cause this or how to re-enable it.
Can anyone tell me how to re-enble mating?

Are you editing a part in the assembly?
I notice this when parts and assemblies are loaded lightweight. They must be resolved to update proper.
You also can't do mates if working in an assy in an exploded mode. Collapse it to make it available.
